Effect of Dietary Fats on Colon Carcinogenesis and Lipid Peroxidation Status in N-Methyl-N¡¯-nitro-N-nitrosoguanidine(MNNG)-Treated Rats

Abstract
The effects of the type of dietary fats and N-methyl-N,-nitro-N- nitrosoguanidine(MNNG) treatment on the status of lipid peroxidation were examined in male spragne Dawley rats. At 8 wks old all rats were given intrarectal injections of saline or MNNG (2 §·/rat) twice a week for 3 wks. All rats were fed for 37 wks one of four diets containing 9% (w/w) fat: BT (7.2% Beef tallow +1.8% corn oil), CO (9.0% cornoil), PO (9.0% perilla oil), FO(2.5% corn oil + 6.5% fish oil). There were no tumors in the control groups but in the MNNG treated rats, the incidence of colonic tumor were 33.3%(l0/30) in the BT+ MNNG group, 14.5%(8/55) in the CO+ MNNG group,10%(3/30) in the PO+ MNNG group, 6.7%(2/30) in the Fo+ MNNG group. Plasma tocopherol and MDA level were significantly reduced in FO+ MNNG group, RBC tocopherol and MDA level were no significant difference. MDA level in liver was significantly higher in BT+ MNNG group and lower in PO+ MNNG group. The activities of SOD and GSH-Px in liver tissue were not affected by the type of dietary fats in controls. However, with MNNG treatment, the level of SOD was significantly increased in CO + MNNG groups and GSH-Px lowered in BT+ MNNG and CO + MNNG groups. In hepatic lipid composition, the relative wt %. of arachidonic acid (AA, c20:4) in the BT and CO groups, ¥á-linolenic acid (c18 : 3) in the PO group, and EPA (c20 : 5) in PO and FO groups was increased significantly, overall, with MNNG treatment, the status of lipid peroxidation of BT+ MNNG group (high hepatic AA level, low EPA level, low GSH-Px level, and high MDA content) could be related to the higher incidence of colon tumor in BT+ MNNG group than the other groups.
